In Gram-negative bacteria, newly made lipopolysaccharide (LPS) is exported to the outer membrane. We report where key steps in the LPS biosynthesis occur in the asymmetric bacterium Brucella abortus. Moreover, we discovered an enzyme with two activities allowing it to complete LPS biosynthesis.
